Perbandingan PERBANDINGAN KUALITAS PREPARAT SITOPATOLOGI CAIRAN EFUSI PLEURA DENGAN PEWARNAAN GIEMSA DAN PAPANICOLAU

ABSTRAK
Pemeriksaan sitopatologi berperan penting dalam menunjang diagnosis berbagai kondisi patologis, terutama pada kasus efusi pleura, dimana kualitas preparat sangat dipengaruhi oleh metode pewarnaan yang digunakan. Penelitian ini bertujuan untuk membandingkan kualitas preparat sitopatologi cairan efusi pleura menggunakan metode pewarnaan giemsa dan papanicolaou. Studi kuantitatif komparatif dengan desain post-test only dilakukan pada 52 sampel efusi pleura yang diperoleh melalui pengambilan sampel total dari pasien yang menjalani torakosentesis di Rumah Sakit Umum Undata Palu, pada Juli 2025. Setiap sampel dipreparasi menggunakan kedua teknik pewarnaan dan dievaluasi berdasarkan morfologi sel, kontras inti, kontras sitoplasma, dan kebersihan latar belakang. Uji Mann–Whitney U menunjukkan perbedaan signifikan antara kedua metode (p < 0,001), yang mengindikasikan bahwa pewarnaan papanicolaou memberikan kontras inti dan sitoplasma yang lebih unggul.
ABSTRACT
Cytopathological examination plays an important role in supporting the diagnosis of various pathological conditions, particularly in cases of pleural effusion, where the quality of the preparations is greatly influenced by the staining method used. This study aimed to compare the quality of cytopathological preparations of pleural effusion fluid using giemsa and papanicolaou staining methods. A comparative quantitative study with a post-test only design was conducted on 52 pleural effusion samples obtained through total sampling from patients undergoing thoracentesis at Undata General Hospital Palu, in July 2025. Each sample was prepared using both staining techniques and evaluated based on cell morphology, nuclear contrast, cytoplasmic contrast, and background cleanliness. The Mann–Whitney U test showed a significant difference between the two methods (p < 0.001), indicating that papanicolaou staining provides superior nuclear and cytoplasmic contrast.
